(1) Tail tip blood collection: This method is used when the required blood volume is small. Fix the animal and trace the mouse tail, soak the mouse tail in warm water at 45 ° C for several minutes, or rub it with chemical drugs such as xylene to expand the local new blood vessels. Dry the mouse tail, cut off the tail tip, and the blood flows out of the tail tip, let the blood drip into the receptacle or directly suck with a pipette. If you need to take a certain period of time, take the blood from the tip of the rat's tail several times. When you take blood, cut off a small section of the tail. After taking the blood, first use a cotton ball to stop the bleeding and immediately apply 6% liquid collodion to At the wound of the tail, a layer of collodion film is formed outside the wound to protect the wound. Blood can also be cut by cutting the tail vein. Three tail potential veins can be cut alternately and cut from the tail tip to the tail root. 0.2 ~ 0.3ml blood can be taken each time. After cutting, the cotton is used to stop bleeding. This method of blood collection is better in rats, and continuous blood can be taken at longer intervals for routine blood tests.


(2) Blood collection from the posterior orbital venous plexus: This method is used when a moderate amount of blood is needed and the animal is prevented from dying. Fix the mouse with the left hand, pinch the skin of the head as much as possible to fix the head, and gently press down on both sides of the neck, causing difficulty in returning blood to the head veins, making the eyeballs fully extrude (showing congestion of the posterior venous plexus), Hold a capillary glass tube and pierce the throat along the posterior wall of the inner palate orbit toward the throat. The penetration depth is 2 to 3 mm in mice and 4 to 5 mm in rats. When you feel resistance, back up later, keep the level, and attract a little. Because of the blood pressure, the blood flows into the glass tube. After obtaining the required amount of blood, pull out the capillary. If the technique is proper, mice can collect about 0.2-0.3ml of blood, and rats can collect about 0.4-0.6ml of blood.


(3) Decapitation blood collection: This method is used when a large amount of blood is needed without further saving the animal's life. Grasp the animal with your left hand and tilt your head slightly downward, and use your right hand to vigorously cut off the mouse head to allow blood to drip into the container. Blood can be collected from 0.8 to 1.0 ml in mice, and 5 to 8 ml in rats.


(4) Orbital artery and orbital vein blood collection: This method can take a large amount of blood, and can avoid the hemolysis caused by the intermixing of tissue fluid in the decapitation blood collection method. It is often used instead of the decapitation blood collection method. After the animal's eyeballs were protruded and congested, the eyeballs were quickly clamped with elbow ophthalmic forceps, and the mouse was turned upside down with the head down, and blood quickly flowed out of the orbit, allowing the blood to drip into the receptacle until it stopped flowing. This method is because the animal is not dead and the heart is beating continuously during the blood collection process, so the blood volume is more than that of the decapitation method. Generally, it can take 4 to 5% of the body weight of the mouse, which is a better method of blood collection.

(5) Blood extraction from the heart: Animals are supine and fixed on a fixed plate, the coat on the anterior heart area is cut off, and the skin is disinfected with iodine alcohol. In the 3rd to 4th intercostal space on the left, use your left index finger to touch the heart beat, and take a syringe with a 4 to 5 gauge needle in your right hand, and choose the puncture where the heart beats the most. When the needle penetrates the heart, the blood will beat because of the heart Power into the syringe automatically. This method requires the experimenter to grasp the following points: insert the heart quickly and directly, otherwise, the heart will slip off from the needle tip; if the needle is not punctured for the first time, remove the needle from the spine and avoid probing around the heart, so as not to damage the heart, Lungs; suck slowly and steadily, otherwise, too much vacuum will collapse the heart. If you don't need to save the life of the animal, you can also numb the animal's chest after intoxication, and pierce the syringe directly into the human heart to suck blood.


(6) Blood extraction from large blood vessels: large and mice can also take blood from cervical arteries, veins, femoral arteries, veins, and axillary arteries and veins. Blood extraction in these areas requires anaesthesia. * Animals are fixed after intoxication, and then operated. After the vein is separated and the exposure is clear, use a syringe to pierce the large blood vessels in parallel (or cut the large blood vessels directly with scissors) to draw the required blood volume. When cutting off the arteries, prevent blood spatter.
